What Brightening Actually Looks Like, Week by Week

Most brightening products work on one pathway. When a serum addresses all five stages of pigmentation, results follow a specific, cumulative pattern. Dr Alison Jamieson explains the 12-week timeline.

If you have followed this series through the five stages of pigmentation, the mechanism of each active ingredient, and the clinical evidence behind them, you are already better informed than most people who have been buying brightening products for years.

But there is one question we have not yet answered directly: what does the correction process actually feel like, week by week, when a serum addresses all five stages simultaneously?

This matters. Because the single most common reason people abandon a brightening protocol is not that the product failed — it is that they expected the wrong things at the wrong time. They see nothing at week two and assume nothing is happening. Or they see rapid improvement in week three and mistake it for the final result, not realising the deeper correction has barely begun.

Weeks one and two — the surface clears

The first changes you will notice have nothing to do with melanin regulation. They come from the alpha-hydroxy acid blend in the formulation: glycolic, lactic, malic, and tartaric acids.

These acids dissolve the bonds between dead, pigmented keratinocytes at the skin's surface. Glycolic acid, at just 76 Daltons, is the smallest AHA molecule and penetrates the epidermis more readily than larger acids. The result is accelerated shedding of the outermost cell layer — the layer where old pigmentation sits most visibly.

What you will notice: skin feels smoother under your fingers. Your morning complexion looks brighter, not because the underlying pigmentation has changed, but because the dull, pigmented surface cells have been cleared more efficiently. Texture improves before tone does.

This is not the serum "working." This is the serum's first mechanism operating at the speed biology allows. The deeper correction is already underway — you simply cannot see it yet.

Weeks two to four — the delivery trucks slow down

Niacinamide, present in the Brightening Serum at a clinically relevant concentration, operates on a different pathway entirely. Rather than reducing melanin production, it interrupts the transfer of melanin from melanocytes to keratinocytes — the process we described in an earlier post as intercepting the delivery trucks.

Published research indicates niacinamide can reduce melanosome transfer by 35 to 68 per cent without affecting melanin synthesis itself. The pigment still exists inside the melanocytes; it simply reaches the skin's surface in smaller quantities.

Between weeks two and four, this mechanism becomes visible. The overall tone of your skin starts to even out. Superficial pigmentation — recent sun spots, mild post-inflammatory marks — begins to fade noticeably. This is often the point where people feel the serum is "working," and it is. But only two of the five pathways are producing visible results so far.

Weeks four to eight — the production orders drop

This is where a multi-pathway serum separates itself from single-ingredient brightening products, and it is the phase most people never reach because they have already switched to something else.

Oligopeptide-68 — the peptide-based active that targets MITF, the master transcription factor controlling melanin production — operates on a longer biological timeline than AHAs or niacinamide. It does not dissolve surface cells or block transfer. It reduces the production signal itself, upstream of everything else. Fewer orders issued to the enzyme floor means less melanin manufactured at every subsequent stage.

In a 12-week clinical trial, Oligopeptide-68 was demonstrated to be more effective than 4 per cent hydroquinone at reducing pigmentation, without the safety concerns that have led to hydroquinone being restricted in Australia, the European Union, and Japan. But the key word is "12-week." This peptide needs time. Its mechanism is regulatory, not ablative — it changes the instructions your melanocytes follow, which means the effect is cumulative and progressive.

Between weeks four and eight, deeper pigmentation that did not respond to surface exfoliation or melanosome interruption begins to lighten meaningfully. In clinical testing, 87 per cent of participants reported more uniform skin tone within this window.

Meanwhile, ascorbic acid and Glycyrrhiza Glabra (licorice) root extract have been quietly inhibiting tyrosinase — the enzyme that converts tyrosine into melanin — since day one. Their contribution is subtle on its own. But when combined with reduced MITF signalling and lower melanosome transfer, the cumulative effect becomes obvious. Each pathway amplifies the others.

Weeks eight to twelve — the compound result

The final phase is not a new mechanism switching on. It is the full compound effect of all five pathways operating simultaneously over a sustained period.

Surface cells are turning over faster, courtesy of the AHA blend. Melanin transfer to the surface is reduced by niacinamide. Production orders from MITF are suppressed by Oligopeptide-68. Tyrosinase activity is inhibited by ascorbic acid and licorice root extract. And — if you have been following the AM Protocol with the Day Cream SPF 15 — the UV trigger that restarts the entire cycle each morning has been interrupted daily.

This is the window where the Oligopeptide-68 12-week clinical trial protocol reached peak measured results. Established pigmentation, sun damage, and uneven tone show their most significant visible correction here.

The distinction is important: most people who describe a brightening product as "not working" abandoned it during weeks two to four, well before the deeper regulatory mechanisms had time to produce visible change. A single-pathway product peaks early and plateaus. A multi-pathway formulation follows a different curve — slower to start, steeper to finish.

Why this timeline depends on SPF

Everything described above assumes one non-negotiable companion: daily broad-spectrum sun protection.

UV radiation is the primary activator of MITF. Every morning your skin meets sunlight without protection, MITF receives a fresh production order — and your evening brightening serum has to counteract it the following night. You are correcting existing pigmentation and creating new pigmentation in a 24-hour loop.

Patience is a protocol decision

The most practical thing this timeline teaches is that the speed of visible results is not determined by how much product you apply or how many days in a row you use it. It is determined by the biology of each pathway, and no ingredient — regardless of concentration — can override the pace at which melanocytes adjust their behaviour.

What a well-formulated multi-pathway serum does is ensure that when each pathway reaches its effective window, every other pathway is already contributing. The result is not one improvement followed by a plateau. It is a series of compounding improvements, each one more visible than the last, reaching their collective peak around the 12-week mark.

If you are two weeks into a brightening protocol and the only change you can see is smoother texture, that is not a failure. That is the AHAs doing their job first, while the rest of the formulation works on the stages you cannot see yet. Stay with the protocol.
