Five Stages of Pigmentation — And Why Brightening Without SPF Is Working Against Yourself
Abstract
If you have been following this series, you now understand something most people never learn about their own skin: pigmentation is not one problem. It is five separate processes, running simultaneously, each one feeding the next.
You know that MITF issues the production orders. That tyrosinase runs the enzyme floor. That melanosomes package and deliver the finished pigment. That niacinamide intercepts the delivery trucks rather than shutting down the factory. That Oligopeptide-68 outperformed 4% hydroquinone in clinical comparison by working upstream, at the production order stage, rather than waiting for melanin to appear and trying to chase it.
But knowing how pigmentation works and knowing what to do about it are two different things. This is the piece that connects the science to the protocol — the part where understanding becomes a routine you can actually follow.
The five stages, mapped to what sits on your bathroom shelf
Here is the pigmentation pathway, simplified to its essentials, with the specific ingredients in the Aliangé Brightening Serum and Day Cream SPF 15 that address each stage.
Stage 1 — The production order (MITF activation)
UV, hormones, and inflammation activate MITF, which tells your melanocytes to ramp up melanin-synthesising enzymes. Oligopeptide-68 in the Brightening Serum downregulates this signal. Fewer production orders issued means less melanin manufactured at every stage below.
Stage 2 — The enzyme floor (tyrosinase activity)
Tyrosinase converts tyrosine into melanin through a chain of oxidation reactions. Ascorbic acid and Glycyrrhiza Glabra (licorice) root extract in the Brightening Serum inhibit this enzyme directly. But they work far more effectively when stage one is already dialled down — which is why single-ingredient brightening products plateau.
Stage 3 — The delivery network (melanosome transfer)
Melanosomes carry finished melanin from melanocytes to keratinocytes via dendrite projections. Niacinamide — present in both the Brightening Serum and the Biocinamide — reduces this transfer by 35 to 68 per cent without affecting melanin synthesis itself. The pigment still exists; it simply does not reach the surface in the same quantity.
Stage 4 — The surface layer (pigmented cell accumulation)
Melanin-loaded keratinocytes stack up at the skin's surface, creating visible discolouration. A blend of alpha-hydroxy acids in the Brightening Serum — lactic, glycolic, malic, tartaric, and citric — accelerates the natural shedding of these pigmented cells. The Transforming Exfoliator, used two to three times per week, supports this turnover further.
Stage 5 — The trigger (UV re-activation)
And here is where most brightening routines quietly fail. Stages one through four can be addressed perfectly, and it will not matter if the trigger that started the whole process keeps firing. UV radiation is the single largest driver of MITF activation. Every time unprotected skin meets sunlight, the factory receives a fresh production order. You are correcting existing pigmentation with one hand and creating new pigmentation with the other.
This is why the Day Cream SPF 15 is not an optional extra in the protocol. It is the stage-five intervention. Without it, stages one through four are running uphill.
The Nambour evidence
In 1992, researchers in Nambour, Queensland — roughly ninety minutes north of Brisbane — enrolled 903 adults in a randomised controlled trial to test whether daily sunscreen use could measurably prevent skin ageing. The study ran for four and a half years. The results, published in the Annals of Internal Medicine in 2013, found that participants who applied sunscreen daily showed no detectable increase in skin ageing over the study period, while the control group aged measurably. The daily-use group experienced 24 per cent less skin ageing than those who used sunscreen at their own discretion.
This was not a laboratory study on isolated cells. It was conducted on real skin, in real Queensland conditions, over years. And the sunscreen used was SPF 15 — the same protection level in the Aliangé Day Cream.
The clinical relevance for pigmentation is direct. UV radiation does not merely age your skin. It activates the MITF pathway that drives melanin production. Every day without broad-spectrum protection is a day your skin receives fresh instructions to produce more pigment — instructions that your evening brightening serum then has to counteract the following night.
The protocol: how this maps to AM and PM

The sequence matters. Brightening actives like AHAs and vitamin C increase photosensitivity, which means skin treated with these ingredients in the evening is more vulnerable to UV the following day. Applying the Brightening Serum at night and following with SPF in the morning is not a preference — it is a clinical sequencing decision.
Why one without the other does not work
There is a common pattern Dr Jamieson observes in patients who report that their brightening products "stopped working." They invested in correction — a well-formulated serum, consistent evening application, patience — and saw initial improvement. Then progress stalled. Then the pigmentation returned.
In almost every case, the missing variable is the same. No consistent daily SPF. The serum was correcting existing pigmentation beautifully, but unprotected UV exposure was generating new pigmentation just as fast. The net result: a plateau that feels like the product has failed, when in reality the product was never given a fair chance to work.
