If you have ever used a brightening serum and felt like it only did half the job, there is a good reason for that. It probably did.

Most brightening products work on a single enzyme — tyrosinase — which is involved in the production of melanin. And they do that reasonably well. But tyrosinase is not the whole story. Your skin actually produces pigment through two separate systems, and unless a formulation addresses both of them at their shared control point, you are only ever managing one half of the equation.

Understanding the difference between these two systems, and where they converge, is the key to understanding why some brightening approaches produce visible, lasting results and others plateau.

Two Systems, One Problem

Your skin manages pigmentation through two distinct mechanisms.

The first is constitutive pigmentation. This is your baseline skin tone — the pigment level determined by your genetics. It is always present, always producing melanin at a steady rate, regardless of sun exposure or environmental triggers.

The second is facultative pigmentation. This is the reactive system — the one that ramps up melanin production in response to UV exposure, hormonal shifts, inflammation, or injury. It is the system responsible for sun spots, post-inflammatory hyperpigmentation, melasma, and the uneven tone that accumulates over years of living under the Queensland sun.

Both systems are real. Both produce melanin. And both contribute to the visible pigmentation concerns that bring people to a dermal clinician's office.

The problem is that most conventional brightening ingredients — vitamin C, licorice extract, arbutin — work downstream, at the tyrosinase level. They inhibit the enzyme that converts tyrosine into melanin. This is useful. But tyrosinase is only one of several enzymes involved, and it sits relatively late in the production chain. Blocking it is a bit like trying to reduce output at a factory by slowing down one machine on the assembly line — the orders are still coming in, the production schedule has not changed, and the factory will find a way to compensate.

To change the production schedule itself, you need to go upstream.

The Master Switch: MITF

At the top of the melanin production pathway sits a transcription factor called MITF — Microphthalmia-associated Transcription Factor. If pigmentation is a factory, MITF is the manager who decides how much melanin gets made. It controls the expression of tyrosinase, yes, but also TRP-1 and TRP-2 — the full suite of enzymes responsible for melanin synthesis.

When MITF is active, the entire production chain runs. When it is downregulated, the factory slows across every stage.

This is where the distinction matters clinically. An ingredient that inhibits tyrosinase alone is working at stage two of a five-stage process. An ingredient that downregulates MITF is working at stage one — it dials down the signal that drives all subsequent stages, in both the constitutive and facultative systems simultaneously.

This is why Dr Jamieson chose Oligopeptide-68 as the lead active in the Brightening Serum.

How Oligopeptide-68 Works

Oligopeptide-68 is a biomimetic peptide that mimics the activity of TGF-β (Transforming Growth Factor-beta), a signalling molecule involved in melanocyte regulation. By mimicking this signal, it downregulates MITF at the transcription level — suppressing the expression of tyrosinase, TRP-1, and TRP-2 simultaneously.

In practical terms, this means the peptide does not simply slow down one enzyme. It reduces the cell's instruction to produce melanin-synthesising enzymes in the first place. It operates upstream of every other brightening ingredient in the formulation.

Published clinical data supports the approach. A 2016 randomised, double-blind trial (Pratchyapurit et al.) compared an Oligopeptide-68 formulation against 2% and 4% hydroquinone over 12 weeks in 38 patients with hyperpigmentation. The peptide formulation matched or outperformed both hydroquinone concentrations, with 76.3% of participants showing moderate improvement — and without the adverse effects associated with long-term hydroquinone use.

That last point is worth noting. Hydroquinone is banned for over-the-counter sale in Australia, the European Union, and Japan due to risks including ochronosis (a paradoxical darkening of the skin) and rebound hyperpigmentation on discontinuation. Oligopeptide-68 offers a different risk profile entirely.

Five Stages, One Serum

Dr Jamieson did not formulate the Brightening Serum around a single active. She built it as a multi-pathway system — one that addresses pigmentation at every stage of the production and delivery process.

Think of it as five points of intervention along the same assembly line.

Stage one — the master switch. Oligopeptide-68 downregulates MITF, reducing the cell's instruction to produce melanin-synthesising enzymes. This is the upstream control point that most brightening products miss entirely.

Stage two — the enzyme factory. Ascorbic acid (vitamin C) and Glycyrrhiza Glabra (licorice) root extract inhibit tyrosinase directly, slowing the conversion of tyrosine to melanin at the enzymatic level.

Stage three — the delivery system. Niacinamide interrupts the transfer of melanosomes — the pigment-carrying packages — from melanocytes to surrounding keratinocytes. Even if melanin is produced, less of it reaches the visible surface of the skin.

Stage four — surface renewal. A blend of alpha-hydroxy acids — glycolic, lactic, malic, tartaric, and citric — accelerates the turnover of pigmented surface cells, revealing less pigmented skin beneath.

Stage five — oxidative defence. Ascorbic acid also functions as an antioxidant, neutralising the free radicals generated by UV exposure that trigger facultative pigmentation in the first place.

Each stage reinforces the others. The upstream MITF suppression means less melanin is produced. The downstream inhibition, transfer interruption, and surface renewal mean less of what is produced ever becomes visible. And the antioxidant defence reduces the environmental trigger that starts the cycle again.

This is protocol thinking applied to a single product — the same logic that governs the broader AM Protocol and PM Protocol.

What This Means in Practice

If you have been using a single-active brightening product — a standalone vitamin C, for instance, or an over-the-counter niacinamide — and the results have plateaued, the reason is likely structural. You are addressing one or two stages of a five-stage process. The factory is still running.
